The Enhanced DAB Peroxidase Substrate Kit is used to detect peroxidase (HRP) enzyme in immunohistochemistry (IHC) staining and immunoblotting.
The Enhanced DAB Peroxidase Substrate Kit consists of a liquid stable DAB chromogen and enhanced DAB buffer. Just before staining, add one drop (~32 µL) of DAB Chromogen for every 1 mL of Enhanced DAB Buffer. The mixed DAB working solution is stable for 4 weeks at 4 ºC in the dark, making it ideal for use in automated stainers. In the presence of peroxidase (HRP) enzyme, the DAB substrate solution produces a brown reaction product.
The Enhanced DAB Peroxidase Substrate Kit can be used on tissue sections or cells, or on membranes such as nitrocellulose, PVDF, or nylon.
Preparation of DAB Working Solution
Add one drop (~32 µL) of DAB Chromogen for every 1 mL of Enhanced DAB Buffer. The mixed DAB working solution can be stored at 4 ºC for 4 weeks.
Instructions For Use
For Tissues or Cells
After incubation with a peroxidase (HRP) detection system, rinse well. Incubate with the DAB working solution (~100 µL) at room temperature for 2-10 minutes or until the desired color intensity is obtained.
Wash for 5 minutes in water.
For Blots
Development time is generally 10-20 minutes at room temperature. When development is satisfactory, rinse membrane in water and air dry.
