Measure Cell Viability and Proliferation based on Metabolic Activity

Introduction

Our Cell-Quant™ MTT Cell Proliferation Assay Kit (A015) and Cell-Quant™ AlamarBlue Cell Viability Reagent (A016) utilize metabolic indicators to measure cellular reduction potential. In the MTT assay, the MTT reagent is reduced by metabolically active cells to a strongly colored reporter, formazan. The signal is proportional to the number of live cells, thus allowing an accurate quantification of changes in the rate of cell proliferation. Similarly, our AlamarBlue reagent (resazurin) is converted to the fluorescent and colorimetric reporter molecule resorufin in metabolically active cells. The fluorescent and colorimetric signal generated from the assay is proportional to the number of living cells in the sample (Figure 1).


Figure 1. Jurkat cells were diluted to the indicated cell numbers in 100 µL volumes, delivered to the wells of a microplate, and incubated for 4 hr with Cell-Quant™ MTT Cell Proliferation Assay Kit, absorbance was measured at 570 nm (Left). In parallel experiment, the cells were incubated with AlamarBlue reagent for 4 hr, fluorescence was measured at Ex/Em=545/590 nm (right).
